Where to invest the next dollar

Priority 1 — Donson 2022 Visium
Confirms or refutes the PFA1 niche insight at spatial resolution. Parser is already written + unit-tested. Acquisition is GEO + email to Donson lab. Cheapest decisive move.
Priority 2 — KEGG/Reactome neural-dev signatures
Sharpens the falsification framework verdict. Hallmark-pathway refutation may be a feature-space artifact; a neural-development-specific lens could give a different answer.
Priority 3 — Raw RNA per sample
Unlocks both the ZFTA-RELA and MERTK insights. Higher cost (significant pipeline rework). Worth doing if those insights matter for separate reasons, not just to unblock them.

What the wiring means

  • Solid arrow (gates) — the source was supposed to gate the target. The falsification gate covered the PFA CDK4/6, ZFTA-RELA, and MERTK insights under the developmental-origin framework. Since the falsification test is now refuted, those gates no longer constrain the targets — but the targets also lose their mechanism story.
  • Dashed arrow (unblocks) — adding the dataset/feature would un-park the target insight. None of these are currently in the ETL pipeline.
  • The PFA1 niche insight has no gating edge in — it's niche-based, not developmental. That's why it's the cleanest path forward.
